1.如果是有Be动词情况下 

Is he not a rich man?  （正确）

Isn't he a rich man? （正确）

 2.如果没有Be动词的情况下

 Did he not come yesterday?  （正确）

Didn't he not come yesterday? （错误）

Didn't he come yesterday? （正确）

 两组句子在表达意思上没有多大差别。

以下两组句子，都正确，但意思差别很大。

Is he not a rich man?  （一般疑问句，说话人不清楚他不是有钱人。）

Isn't he a rich man? （修辞反问句，表示更加肯定的语气：难道他不是有钱人吗？）

Did he not come yesterday? （一般疑问句，说话人不清楚他昨天没来。）

Didn't he come yesterday? （这句估计多写了一个 not，去掉。修辞反问句，表示更加肯定的语气：难道他昨天没来吗？）

意义上没有差异，但语体差异是很大的。正式语体从不用not与操作词的缩合形式。isn't, doesn't都是非正式体。Is sb not...? Does sb not...? 为正式语体。
